Total nitrogen determination

چکیده

As a preliminary to the consideration of automated, or more correctly mechanized, methods for the determination of total organic nitrogen the discussion of some general aspects of nitrogen determination is illuminating. There are at least four recent reviews, for example those by Bradstreet (1965), Fleck & Munro (I965), Jacobs (1965), and Munro & Fleck (1969), of the determination of nitrogen from which certain general principles for the estimation of nitrogen in samples of biological materials can be derived. An up-to-date review of analytical methods is incomplete without the mention of quality control. It is now apparent in the field of clinical biochemistry that continuous and daily checks of analytical precision and accuracy are essential for results to be reliable; the determination of nitrogen is no exception. The two principal methods of determining organic nitrogen were first described almost a century ago, the Kjeldahl method in 1883 and the Dumas method in 1831, yet it is only in recent years that important alterations in techniques have led to considerable improvements. The advent of gas-liquid chromatography was followed by considerable refinements in the Dumas method so that it is now possible to analyse dry solid samples rapidly, only a few minutes being required for each determination of nitrogen (see Fleck & Munro, 1965). The Kjeldahl method, however, remains the most widely used and convenient for samples of biological origin. The subsequent discussion relates only to the determination of the nitrogen content of samples of animal tissues, excreta and food. Substances such as keratin, horn, or soils may present difficulties and for such special applications it is essential to consult the recent reviews and to carry out careful recovery experiments with the technique adopted.

Determination of total nitrogen in aqueous samples using persulfate digestion

Determination of total nitrogen in aqueous samples after potassium persulfate digestion compared favorably in both precision and nitrogen recovery with determinations obtained using Kjeldahl digestion. The determination of total nitrogen (TN = inorganic and organic fixed nitrogen) has largely depended either on acid Kjeldahl digestion ( cf. Strickland and Parsons 1972) or on photo-oxidation (Ar...

A Rapid Automated System for the Analysis of Dissolved Total Organic Nitrogen in Aqueous Solutions

-An automated system for the analysis of dissolved organic nitrogen has been developed and applied to natural waters. It is based on subjecting sample to u.v. irradiation followed by a heterogeneous reduction of the nitrogen containing irradiation products to ammonia which is detected by an ammonia gas sensing probe. Quantitative recoveries of different organic nitrogen compounds common to natu...

Nondestructive Determination of the Total Volatile Basic Nitrogen (TVB-N) Content Using hyperspectral Imaging in Japanese Threadfin Bream (Nemipterusjaponicus) Fillet

Background and Objectives: Considering the importance of safety evaluation of fish and seafood from capture to purchase, rapid and nondestructive methods are in urgent need for seafood industry. This study aimed to assess the application of hyperspectral imaging (HSI: 430-1010 nm) for prediction of total volatile basic nitrogen (TVB-N) in Japanese-threadfin bream (Nemipterusjaponicus) fillets, ...

Methods for the Determination of Submicro Quantities of Total Nitrogen, Ammonia, Amino Nitrogen, Amides, Peptides, Adenylic Acid, and Nitrates

In 1935 one of us described micromethods for ammonia, urea, total nitrogen, uric acid, creatinine, and allantoin (1). These methods are adapted for the analysis of 1 to 2 ml. aliquots of dilute solutions, e.g. 0.1 mg. per cent of ammonia or urea, and for carrying out a large number of analyses simultaneously. These methods all end in a calorimetric measurement. In the present communication meth...
